The microtiter plate provided in this kit has been pre-coated with 5-HIAA / 5-Hydroxyindoleacetic Acid. During the reaction, 5-HIAA / 5-Hydroxyindoleacetic Acid in the sample or standard competes with a fixed amount of 5-HIAA / 5-Hydroxyindoleacetic Acid on the solid phase supporter for sites on the Biotinylated Detection Antibody specific to 5-HIAA / 5-Hydroxyindoleacetic Acid. Excess conjugate and unbound sample or standard are washed from the plate, and HRP-Streptavidin(SABC) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. The concentration of 5-HIAA / 5-Hydroxyindoleacetic Acid in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Competitive ELISA Protocol
The below protocol is a sample protocol for a competitive ELISA kits. Competitive ELISA kits allow for the detection and quantification of an analyte in a sample. This allows the researcher to calculate the amount of analyte present in their sample. This can be very useful when looking for increases in protein concentration, phosphorylation of proteins or decreases on protein concentrations.Before adding to wells, equilibrate the SABC working solution and TMB substrate for at least 30 min at 37 °C. When diluting samples and reagents, they must be mixed completely and evenly. It is recommended to plot a standard curve for each test.
